چکیده

This presentation describes the events that led to the sequential discoveries of nerve growth factor (NGF) and epidermal growth factor (EGF). Each was isolated during attempts to understand the biochemical basis for unexpected biological observations. The in vivo stimulation of the growth of specific nerve fibers following the transplantation of a mouse sarcoma into a chick embryo eventually resulted in the finding of NGF, not only in the transplanted tumor but also in snake venom and in the mouse salivary gland. Unexpectedly, treatment of newborn animals with extracts of the mouse salivary gland resulted not only in enhancement of nerve growth but also in precocious eyelid opening and tooth eruption. The latter observations led to the isolation of both EGF and its tyrosine kinase-active receptor.

Ionic Responses and Growth Stimulation Induced Nerve Growth Factor and Epidermal Growth Factor in Rat Pheochromocytoma (PC12) Cells

Rat pheochromocytoma cells (clone PC12) respond to nerve growth factor (NGF) by the acquirement of a phenotype resembling neuronal cells. In an earlier study we showed that NGF causes an increase in Na+,K + pump activity, as monitored by ouabain-sensitive Rb + influx. Here we show that addition of epidermal growth factor (EGF) to PC12 cells resulted in a stimulation of Na+,K ÷ pump activity as ...

Comparison of rapid changes in surface morphology and coated pit formation of PC12 cells in response to nerve growth factor, epidermal growth factor, and dibutyryl cyclic AMP

Scanning and transmission electron microscopic studies were carried out on the rapid cell surface response of PC12 pheochromocytoma cells to treatment with nerve growth factor (NGF), epidermal growth factor (EGF), and dibutyryl cyclic AMP. EGF induced a rapidly initiated series of surface changes identical to those previously observed with NGF. Ruffles appear over the dorsal surface of the cell...

Glucose and amino acids modulate translation factor activation by growth factors in PC12 cells.

In PC12 phaeochromocytoma cells, protein synthesis is activated by epidermal and nerve growth factors (EGF and NGF). EGF and NGF also regulate a number of components of the translational machinery in these cells. Here we show that the ability of EGF and NGF to induce the phosphorylation of the 70 kDa ribosomal protein, S6 kinase, and the eukaryotic initiation factor (eIF), 4E-binding protein 1,...

Synthetic chimeras of mouse growth factor-associated glandular kallikreins. II. Growth factor binding properties.

Six chimeric constructs of the sequentially similar growth factor-associated kallikreins-epidermal growth factor binding protein (EGF-BP) and the gamma-subunit of nerve growth factor (gamma-NGF)--have been expressed, and their ability to generate complexes with epidermal growth factor (EGF) and beta-NGF, analogous to the high molecular weight forms (7S NGF and HMW-EGF) found in the mouse submax...

Early changes in protein synthesis induced by basic fibroblast growth factor, nerve growth factor, and epidermal growth factor in PC12 pheochromocytoma cells.

Nerve growth factor (NGF) and basic fibroblast growth factor (bFGF) stimulate neuronal differentiation, whereas epidermal growth factor (EGF) promotes only mitogenic responses in PC12 pheochromocytoma cells. The early changes in protein synthesis induced by bFGF, NGF, and EGF in these cells have been determined by two-dimensional PAGE of [35S]methionine-labeled proteins and computerized image a...
